- Sir Ambrojoe – kill 6,000 Stage Killers in Cappella Magna.
- Red Death – kill death after time runs out (requires both Crimson Shroud and Infinite Corridor weapon evolutions).
- Leda – in Gallo Tower, defeat Leda. In the beginning, turn south until you see Leda. When you get close to it, the screen will go black and the soundtrack will get sinister.
- Boon Marrabbio – in Mad Forest, vanquish the shadow. Get the Skull and Pummarola objects, then continue to collect chickens from braziers until pies start to emerge to make the shadow materialize. Eat pie continuously till the shadow appears. By gathering the Clover, you can enhance the likelihood that a brazier will spawn.
- Minnah Mannarah – the Dairy Plant werewolves must be vanquished. As you begin your run, select Arcana VIII – Mad Groove, and then gather the cheese that appears to summon the werewolves. The new character won’t be unlocked if a werewolf despawns rather than dies.
